Primary Cells

Human telomerase reverse transcriptase (hTERT) immortalized cell lines represent an intracellular biology study that combines the in vivo nature of primary cells with the ability of a conventional cell line to survive in vitro continuously.
Since rapid dilution decreases the viability, it should be diluted slowly after dissolving. Sudden dilution with DMSO causes osmotic shock, decreasing cell survival in half.
The MEM and EMEM formulation are not the same . The biggest difference between the two is in the non-essential amino acids in EMEM. Since ATCC only grows these cells in formulated EMEM, this cell line will be accustomed to EMEM.
Sudden changes in the areas where cells are acclimated can cause cells to go into shock or deteriorate their health. Make sure you always use the recommended media.
Floating cells should not be removed from the parent culture until it is certain that they are dead. This causes the cell density to drop below a critical value and the growth of the cells to be delayed.
The use of heat inactivated FBS when working with cell lines is not recommended unless specifically stated. The reason for this is that the heat inactivation of the serum destroys various cell growth and binding factors.
